熏洗方对大鼠骨折后期僵硬关节滑膜Ⅲ型胶原mRNA及TGFβ_1表达的影响 被引量:24

摘要 为探讨熏洗方治疗大鼠骨折后期关节僵硬的作用机理。以Wistar大鼠制作骨折后期关节僵硬模型 ,用dotblot杂交法和ELISA法分别测定踝关节滑膜Ⅲ型胶原mRNA表达水平及TGF β1含量。结果显示模型组Ⅲ型胶原mRNA及TGFβ1水平明显升高 ,与正常对照组比较差异有显著性 (P <0 .0 1,P <0 .0 5 ) ;功能组和中药组Ⅲ型胶原mRNA、TGFβ1水平明显降低 ,与模型组比较差异有显著性 (P <0 .0 1) ;中药组Ⅲ型胶原mRNA、TGFβ1水平显著低于功能组 (P <0 .0 5 )。表明大鼠骨折后期关节僵硬滑膜Ⅲ型胶原mRNA及TGFβ1水平异常改变 ;降低Ⅲ型胶原mRNA及TGFβ1水平可能是熏洗方治疗骨折后期关节僵硬的重要机制之一。 In order to study the action mechanism of 'XUN XI FANG' (XXF, a TCD recipe for steaming and washing) on collagen-III mRNA expression and TGF-β1 level in stiff joint synovium at the late stage of rat fracture, animal models of stiff joint at the late stage of fracture were made on Wistar rats, the collagen-III mRNA expression and TGF-β1 level in the ankle joints were detected by dot blot hybradization and ELISA methods, respectively. The results showed: (1) the collagen-III mRNA and TGF-β1 levels in the modeled group ascent significantly with a significant difference from those of the normal control group(P < 0.05, P < 0.01); (2) the collagen-III mRNA and TGF-β1 levels in the functional and TCD groups descended obviously with a significant difference from those of the modeled group (P < 0.01); (3) the collagen-III mRNA and TGF-β1 levels of the TCD group were significantly lower than those of the functional group (P < 0.05), suggesting that at the late stage of rat fracture, the collagen-III mRNA and TGF-β1 levels in the stiff joint synovium were abnormally changed; the function to reduce the collagen-III mRNA and TGF-β1 levels may be one of the important mechanisms of XXF for treating stiff joint at the late stage of fracture.
