摘要 目的:探讨白细胞介素 10(IL 10)对大鼠移植肺再 灌注损伤的保护作用及机制.方法:将SD大鼠36只随机分 为3组,每组12只,即假手术对照组(Control)、肺移植组和肺 移植加IL 10处理组.观察受体气管内滴入IL 10对移植肺再 灌注损伤的保护作用.肺功能指标包括血氧分压、肺湿干重 比、肺通透性指数、支气管肺泡灌洗液(BALF)中白细胞计数 和分类;ELISA法检测受体血清中肿瘤坏死因子α(TNFα)含 量.结果:肺移植术后1h,肺湿干重比、肺通透指数和BALF 中中性粒细胞百分比分别为(6.57±0.89)、(23.49±5.13)× 10-4和(49.84±6.53)%,显著高于对照组(P<0.01),经气 管内给予受体大鼠IL 10可明显降低上述指标至(4.23± 0.58)、(15.96±3.04)×10-4和(20.33±4.26)%(P< 0.01);肺移植组血清TNFα含量(μg/L)为(0.86±0.11),较 对照组显著升高(P<0.01),IL AIM: To investigate the protective effects of Interleukin-10 (IL-10) on ischemia/reperfusion (I/R)-induced lung injury following rat lung transplantation and to explore its mechanisms. METHODS: Thirty-six SD rats were randomly divided into 3 groups with 12 rats in each group: sham operation group (control), lung transplantation group and IL-10 treatment group. IL-10 was administered to the recipient rats through intra-trachea way. The wet/dry ratio of lung, lung permeability index and neutrophil percentage in bronchoalveolar lavage fluid (BALF) were detected. The serum level of tumor necrosis factor α(TNFα)was detected by enzyme-linked immunosorbent assay (ELISA). RESULTS: The wet/dry ratio of lung, lung permeability index and neutrophils percentage in BALF of I/R group were (6.57±0.89) ,(23.49±5.13)×10 -4 and (49.84±6.53)%, significantly higher than those of the control group(P<0.01). The administration of IL-10 to recipient rats before the operation markedly reduced the indexes above to (4.23±0.58), (15.96±3.04)×10 -4 and (20.33±4.26)%, respectively(P<0.01). The serum level of TNFα(μg/L)increased to (0.86±0.11) after lung transplantation, markedly higher than that of control group, however, the IL-10 treatment could reduce its level significantly to [(0.39±0.05),P<0.01]. CONCLUSION: IL-10 protects the lung function against I/R injury after transplantation, which is related to its inhibition of TNFα release and activation of neutrophils.
