摘要
目的 将生长抑素受体 2 (SST2R)基因转染至胰腺癌细胞株PC 3细胞 ,探索奥曲肽对转染SST2R基因前后PC 3细胞凋亡抑制因子 (Survivin)表达量的调节作用。方法 采用脂质体将SST2R基因转染至PC 3细胞 ,免疫细胞化学检测其转染效果。逆转录 聚合酶链反应 (RT PCR)法半定量检测不同浓度 (0 .10、0 .2 0、0 .40、0 .80mg/L)奥曲肽对PC 3细胞转染SST2R基因前后Survivin表达量的调节。结果 转染SST2R基因PC 3细胞Survivin的表达量在不同浓度奥曲肽 (0 .2 0、0 .40、0 .80mg/L)作用下分别为0 .5 5 2± 0 .0 5 9、0 .465± 0 .0 5 0、0 .3 96± 0 .0 68,未转染SST2R基因PC 3细胞Survivin的表达量在不同浓度奥曲肽 (0 .2 0、0 .40、0 .80mg/L)作用下分别为0 .745± 0 .0 93、0 .667± 0 .0 66、0 .63 7± 0 .0 80 ,相同剂量奥曲肽对转染组PC 3细胞Survivin表达量的下调作用更为明显 (P <0 .0 5 )。结论 生长抑素用于PC 3细胞其效果不佳可能与其SST2R下调有关 ,转染SST2R基因后可增加胰腺癌细胞SST2R的表达量 ,同时下调了Survivin表达量 ,它可增强生长抑素对胰腺癌细胞株杀伤力。
Objective To observe the expression of Survivin after using octreotide on PC-3 cells by transfecting somatostatin type 2 (SST2R) gene into human pancreatic cancer cells (PC-3).Methods SST2R gene was transfected into PC-3 cells by liposome.The result of transfection was detected by immunocytochemical staining.The expression of Survivin in PC-3 cells after using different concentrations of octreotide was semi-quantified by RT-PCR.Results The Survivin expression induced by octreotide in transfected PC-3 cells was 0.552± 0.059 at the dosage of 0.20 mg/L, 0.465± 0.050 at the dosage of 0.40 mg/L and 0.396± 0.068 at the dosage of 0.80 mg/L respectively.Survivin expression induced by octreotide in non-transfected PC-3 cells was 0.745± 0.093 at the dosage of 0.20 mg/L, 0.667± 0.066 at the dosage of 0.40 mg/L and 0.637± 0.080 at the dosage of 0.80 mg/L.Octreotide could decrease the survivin expression in transfected groups as compared with that in non-transfected groups (P< 0.05) at the tested dosages ( 0.20, 0.40 and 0.80 mg/L).Conclusion Deficiency of SST2R is probably the major reason that octreotide takes little effect on pancreatic cancer cells.Transfecting SST2R gene can increase the expression SST2R and decrease the expression of Survivin,both of these can enhance the killing effects of octreotide on PC-3 cells.
