大鼠骨骼肌卫星细胞体外培养的实验研究

Culture of rat musclesatellite cell in vitro

目的 改进鼠肌卫星细胞体外培养方法 ,得到更高纯度的肌卫星细胞。方法 采用成年Wistar大鼠 ,将两步消化法加以改进 ,采用Ficoll分离和差速贴壁法两步纯化得到更高纯度的肌卫星细胞。所得细胞采用免疫组织化学方法加以鉴定。结果 此方法肌卫星细胞纯化率可达到98%。细胞增殖快 ,生长良好。结论 本实验成功建立了卫星细胞纯化方法 ,适用于心外科及相关科室开展细胞移植和基因治疗方面的研究。

Objective To improve the culture method of rat muscle setellite cell in vitro and obtain more purified muscle setellite cells.Methods The more purified satellite cells were obtained from adult Wistar rats isolated by improved two-steps digestion method,and purified by the two-step method of Ficoll seperation and velocity sedimentation.The cells were identified by immunochemical stain.Results The purification rate of satellite cells was 98 % by using this method and they showed strong proliferative ablity and grew well.Conclusion The purification technique for satellite cell was developed successfully.It was useful for cardial surgery department or other departments to study cell transplantion and gene therapy.
