热休克蛋白90在门静脉高压大鼠肠系膜血管中的表达

The expression of HSP90 in the endothelium of mesentery vascular in portal hypertensive rats

目的 研究门静脉高压症时肠系膜血管热休克蛋白 90 (HSP90 )的表达 ,并对其表达定位。方法 SD大鼠分为实验组和假手术组 ,用门静脉部分结扎法制作门静脉高压模型。 4周后取肠系膜血管组织 ,采用免疫组织化学技术、免疫印迹 (Westernblot)、逆转录 聚合酶链反应 (RT PCR)法在蛋白及mRNA水平测定门静脉高压大鼠肠系膜血管HSP90的表达。结果 门静脉高压大鼠肠系膜血管HSP90的表达增强 ,且保持在蛋白及mRNA水平上的一致。HSP90不仅在血管内膜而且还在血管平滑肌层有明显的表达。结论 HSP90可能参与了门静脉高压内脏血管病变的形成 ,从而为进一步揭示门静脉高压内脏血管病变的发病机制提供了新的思路。

Objective To examine Hsp90 expression and localization in rat mesenteric vasculature.Methods Sprague-Dawley rats were inducted portal hypertension by partial portal vein ligated(PPVL).Studies were performed in 24 h fasted rats 28 days after PPVL.Immunohistochemistry and Weatern-Blot were used to examine the expression of HSP90 in mesenteric vasculature at protein level.HSP90 mRNA was detected by RT-PCR.Results The expression of HSP90 is significant enhanced in PVL animals compared with that in sham-operated animals whether by Western blot at protein level or by RT-PCR at mRNA level.The immunohistochemistry shows that HSP90 staining not only occurs in vascular endothelium but also occurs in mesenteric smooth cells.Conclusion This study indicates that HSP90 is overexpression in mesenteric vasculature in portal hypertensive rats.
