摘要
通过细胞计数、蛋白合成率、克隆形成率及培养基中乳酸脱氢酶(LDH)活力测定,观察L-缬氨酸(L-Val)浓度对培养胃癌细胞(MKN_(45))及对照细胞(纤维母细胞IMR-90)的影响,发现将L-Val减少至标准MEM(Minimum Eagle Medium)L-Val浓度(4.6 mg/100 ml)的1/16或1/8时,胃癌细胞计数、蛋白合成率及克唐形成率显著降低;而培养基中LDH活力显著上升(P<0.05),纤维母细胞则无类似变化。L-Val浓度加倍或剥夺MEM中L-亮氨酸或L-蛋氨酸对这两种细胞这些特性均无影响。上述结果为配制胃癌不平衡氨基酸治疗液提供了基础。
To confirm the effects of varying L-valine (L-val) concentrations on gastric cancer cells (MKN45) and control cells (fibroblast IMR-90) in culture, the cell count, the rate of protein synthesis, clonning effecieney, LDH activity in culture medium were determined. The reduc- tion of L-val to 1/16 or 1/8 of 4.6 rag/100 ml in common MEM resulted in a significant dec- rease (P<0.05) in the cell count, the rate of protein synthesis and clonning effeciency but a significant increase (P<0.05) in the LDH activity in the culture medium of gastric cance cells and the fibroblast had no such changes. No obvious influence on changes in the property of gastric cancer cell as well as fibroblast was seen by doubling the L-val content or depri- ving the MEM of. L-leucine or L-methionine. The above result provides some theoratical basis in preparing unbalanced aminoacid solution for the treatment of gastric cancer.
