肺高低转移癌细胞株在Survivin突变基因作用下的细胞凋亡

摘要 目的 :研究SurvivinThr3 4 →Ala突变基因对肺低转移癌 95C和高转移癌 95D细胞株的不同凋亡作用。方法 :构建SurvivinThr3 4 →Ala突变基因 ,分别转染 95C、95D细胞株 ,培养 72h流式细胞仪测凋亡率 ;并且肺癌 95C、95D细胞株分别行Survivin免疫组化染色。结果 :SurvivinThr3 4 →Ala转染 95C、95D后 72h凋亡率分别为 15 .79± 2 .95 % (95C)和 2 7.5 7± 2 .0 5 % (95D) ,95D高于95C ;免疫组化显示 95DSurvivin蛋白表达较 95C高。结论 :SurvivinThr3 4 →Ala对肺癌 95C、95D细胞株具有诱导凋亡作用 ,且Survivin表达高的肺癌细胞比表达低的肺癌细胞效果好。 Objective: To study the different apoptosi s i n lung carcinoma 95C and 95D cell strain caused by Survivin Thr 34→Ala mut ant. Method: The Survivin Thr 34→Ala mutant was introduce d by site-directed mutagenesis into the Survivin cDNA. Lung carcinoma 95C and 9 5D cells were transfected by Survivin Thr 34→Ala mutant and cultured for 7 2 hours. Apoptosis rate was estimated by flow cytometry. Immunohistochemical a ssay for Survivin was also applied. Results: The apoptosis index es in 95C、95D cells transfected by Survivin Thr 34→Ala after 72 hours wer e 15.79±2.95% (95C) and 27.57±2.05% (95D) respectively. The apoptosis index i n 95D is higher than that in 95C. Immunohistochemical assay showed 95D had a hi gher expression of Survivin than 95C. Conclusion: Survivin Thr 34→Ala can both trigger apoptosis in cell lines 95C and 95D, and the apop tosis effect on the lung cancer cells with higher expression of Survivin is bett er than cells with lower Simj urvivin expression.
