透明质酸特异的细胞游走受体和CD_(44)V6在膀胱癌侵袭转移中的作用 被引量:1

摘要 目的 :探讨透明质酸特异的细胞游走受体 (receptorforHA -mediatedmotility ,RHAMM)、CD44V6在膀胱癌侵袭转移中的作用。方法 :采用免疫组化SP法检测 6 4例膀胱癌中RHAMM、CD44V6表达 ;采用Westernblot法观察膀胱癌中RHAMM蛋白表达。结果 :膀胱癌RHAMM、CD44V6阳性表达率分别为 6 0 .9%和 5 3.1%。随着膀胱癌进展RHAMM阳性表达有增高趋势 ,与病理分期、术后复发及转移有关 (P <0 .0 5 ,P <0 .0 5 ,P <0 .0 1) ,而CD44V6阳性表达则显著下降 ,与术后复发及转移呈负相关 (P <0 .0 1,P <0 .0 5 )。Westernblot显示膜型RHAMM蛋白在侵袭转移的膀胱癌中有上调表达。结论 :膜型RHAMM、CD44V6异常表达与膀胱癌侵袭转移及预后有关 ,可能是判断膀胱癌恶性表型及预后的指标之一。 Objective: To explore the role of recep tor for HA-mediated motility (RHAMM) and CD 44V6 in the invasion and metastasis of bladder carcinoma. Methods: The expression of RHAMM an d CD 44V6 in 64 cases of bladder carcinoma were studied by immunohistochemist ry; The level of RHAMM protein in bladder carcinoma was estimated by Western blo t simultaneously. Results: The positive expression rate of R HAMM、CD 44V6 in bladder carcinoma were 60.9% and 53.1%, respectively. With the development of bladder carcinoma, the expression of THAMM increased and wer e related to the pathological stages, postoperational recurrence and metastasis (p<0.05,p>0.05, p<0.01, respectively), while the expression of CD 44V6 decreased sharply and reversely related to the postoperational recurren ce and metastasis (p<0.01, p<0.05, respectively). Western blot suggested that the membrane-type RHAMM protein was up-regulated in the bladder carcinoma with invasion and metastasis. Conclusion: The abnormal exp re ssions of RHAMM and CD 44V6 were related to the invasion, metastasis and pr ognosis of bladder carcinoma, and may be one of the indexes for deciding the mal ignant phenotype and prognosis of bladder carcinoma.
