摘要
目的 观察RGD多肽 (2 2 5 )对卵巢切除 (OVX)小鼠骨密度、骨强度和骨代谢的影响。方法 4 0只平均体重为 2 9g的CD - 1雌性小鼠分为 5组 ,OVX组、腹腔注射E2 组 (15 μg·kg-1·d-1)、RGD多肽组 (每只 35nmol d)、假手术组 (Sham)和基础组 (BL) ,测定各组小鼠的骨密度 (BMD)、骨矿含量 (BMC)、骨生物力学及生化指标并进行比较。结果 OVX组小鼠子宫萎缩 ,骨密度和骨矿含量降低 ,骨生物力学性能下降 ,骨转换增强。RGD多肽 (2 2 5 )组骨密度、矿盐含量显著升高 ,弹性模量和弹性桡度明显上升 ,血清ALP明显下降。结论 RGD多肽 (2 2 5 )能明显提高骨密度 ,增加矿盐含量 ,改善骨的生物力学和抑制骨转换。
Objective This study is to observe the effects of RGD-peptide (225) (RGD) with 17β-Estrodiol (E 2) on bone mass,bone mechanical strength and bone metabolism in ovariectomized (OVX) mice.Methods Bone mineral density (BMD),Bone mineral contend (BMC),bone strength and biochemical markers were measured in each group.45(10-week-old) female CD-1 mice,with an average weight of 29 g,were randomly divided into 5 groups.17β-Estradiol (15μg·kg -1·d -1,injection),RGD-peptide (225) (35 nmol/mouse·d) injection for 7 weeks in ovariectomized mice.And it was compared with sham group and BL group.Results Biomechanical parameters decreased,BMD and BMC decreased obviously,and bone turnover also accelerated in OVX mice,compared with sham operated group.After 7 weeks' treatment,the BMD and BMC of RGD Pdypeptide (225) were obviously increased.A part of biomechanical parameters increased and serum ALP significantly decreased in RGD and E 2 group.Conclusion RGD-peptide (225) may increase BMD,BMC,and improve the bone mechanical strength and inhibit the bone turnover obviously.
