摘要
以'洛阳红'叶柄为实验材料,探讨不同基本培养基(MS、1 2MS、WPM)、抗氧化剂与吸附剂(聚乙烯吡咯烷酮、维生素C、硫代硫酸钠)、暗处理、母株预处理以及选材时期等对褐化防止的有效性。结果表明,以WPM为基本培养基时褐化现象最轻,1 2MS次之,MS褐化现象最为严重;三种抗氧化剂和吸附剂对褐化的抑制作用较为明显,其中以PVP最好,VC较次之,Na2S2O3最差;外植体采取前对母株进行遮光处理以及在培养过程中对材料进行暗处理有利于抑制褐化;培养基中生长调节物质(BA、NAA)的浓度变化对褐化没有明显的影响。
The influence factors on browning of leafstalk in tissure culture of Paeonia suffruticosa is discussed. The results showed that basal medium, adding antioxidant and adsorbing substancecan, total darkness, pretreatmenting maternal and selecting material have effects on the rate of browning. The efficient method of decreasing the rate of browning is using the agar medium containing the mineral salts of WPM and PVP500ppm、keeping in total darkness or pretreatmenting maternal. However, the concentration of 6-BA and NAA have no effect on the rate of browning of Paeonia suffruticosa.
