lmbr1基因剪接方式分析

摘要 目的:探讨lmbr1基因的剪接方式。方法:采用序列比对blast方法,结合基因常见的GU AG剪切方式,对人和鼠c7orf2 /lmbr1基因的内含子 外显子边界进行分析;将从GenBank筛选到的lmbr1cDNA序列及其预测蛋白与其常见转录产物的序列及预测蛋白序列比较,进行lmbr1基因可变剪接的分析。结果:获得了lmbr1基因各个外显子的边界和大小,表明c7orf2 /lmbr1基因有多种转录体,分别预测编码不同长度的蛋白质,其剪接方式包括转录起始位点的改变、外显子丢失、内含子驻留和polyA位点的改变等。结论:lmbr1有多种可变剪接方式,且在不同物种其同源基因有相似的转录产物。 Aim: To explore the splicing form of lmbr1 gene. Methods: Using the blast method combining with the common GU-AG splicing method, the exon-intron boundary of c7orf2/lmbr1 gene in human and mice was analyzed. By comparing several lmbr1 cDNA sequences from GenBank and their predicted protein with those of common transcripts in human and mice, the alternative splicing form of lmbr1 was analyzed.Results: The exon-intron boundary of human and mouse lmbr1 gene was obtained.It was suggested that lmbr1 had multiple transcripts that encoded different proteins with different length.The splicing forms included exon skipping, intron retention, initiation site change, and alternative polyA signal site. Conclusion: lmbr1 should have multiple alternative splicing forms, and there should be similar transcripts in different spieces.
