IL-2与TCRγ独特型DNA疫苗联合诱导小鼠特异性免疫反应

摘要 目的:观察白细胞介素-2(IL-2)与代Rγ独特型DNA疫苗诱导BALB/c小鼠抗肿瘤独特型免疫反应.方法:24只6~8周的健康纯系雄性BALB/c小鼠随机均分为3组,在小鼠双侧股四头肌肌内注射2.5 g/L盐酸布比卡因,每侧50μl,1次/d,5 d后分组免疫.各小鼠分别于第1周、2周、4周各免疫1次,共3次.①A组:每次于股四头肌肌内注射pcDNA3 1 100μg;②B组:每次于股四头肌肌内注射pcDNA3.1-TCRy 100μg;②C组:每次于股四头肌肌内注射pcDNA3.1-TCRγ100μg及IL-2 5μg.于末次免疫接种后第5 d将每组小鼠随机选2只处死,分离出胫前肌,RT-PCR法检测重组质粒在小鼠骨骼肌中的mRNA表达;间接免疫荧光法检测小鼠血清中抗体生成情况.结果:在pcDNA3.1-TCRγ组及pcDNA3.1-TCRγ+IL-2组小鼠中用RT-PCR法均检测到了重组质粒在肌肉中的mRNA表达.pcDNA3.1-TCRy组及pcDNA3.1-TCRγ+IL-2组小鼠血清中均产生了特异性抗独特型抗体.在第6周时,pcDNA3.1-TCRγ组小鼠抗体滴度最高达1:160;pcDNA3.1-TCRγ+IL-2组小鼠抗体的滴度最高达1:640,2组差异有统计学意义(P<0.01).结论:TCRγ表达载体pcDNA3.1-TCRγ可以诱导小鼠产生特异性抗淋巴瘤细胞独特型抗体;应用IL-2免疫佐剂可使TCRy独特型DNA疫苗诱导的抗体滴度明显增高. Aim: To investigate the anti-tumor immuneologic effects induced by IL-2 combined with TCRγ idiotypic DNA vaccine on BALB/c mice. Methods: Twenty-four BALB/c mice were randomly allocated into three groups (eight mice in each group): group pcDNA3.1,group pcDNA3.1-TCRγ,and group pcDNA3.1-TCRγ+IL-2. Vaccines were injected in bilateral quadriceps femorise once at 1st,2nd, and 4th week,respectively.The specific idiotypic antibody was detected by indirect immunofluorescence staining. The mRNA in skeletal muscle cell was detected by RT-PCR. Results: The results showed that the specific idiotypic antibody could be found in group pcDNA3.1-TCRγ and group pcDNA3.1-TCRγ+IL-2. At the same time,the antibody titers were higher in group pcDNA3.1-TCRγ+IL-2 than that of group pcDNA3.1-TCRγ(P<0.01).The highest antibody titer was 1GA6FA640 in the group pcDNA3.1-TCRγ+IL-2, whereas the highest titer was 1GA6FA160 in the group pcDNA3.1-TCRγ.The mRNA expression in skeletal muscle cell could be found in group pcDNA3.1-TCRγ and group pcDNA3.1-TCRγ+IL-2.Conclusion: This recombinant pcDNA3.1-TCRγ could be used as DNA vaccine to induce specific anti-idiotypic antibody against lymphoma. Using IL-2 as immunologic adjuvant could significantly increase the antibody titers.
