摘要 目的 研究实验性糖尿病大鼠心肌细胞凋亡以及凋亡相关蛋白的表达。方法 链脲菌素 (STZ)法复制SD大鼠实验性糖尿病模型。于模型成功后 30、4 5、6 0、75及 90d取材。原位末端标记法 (TUNEL)和免疫组化法检测细胞凋亡及心肌组织中凋亡相关蛋白的表达变化。结果 健康对照大鼠心肌仅有少量凋亡细胞 ,p5 3、Fas和Bcl 2的免疫反应阳性 ,Bax免疫反应阴性。糖尿病 30d后 ,心肌的凋亡细胞阳性指数开始升高 ,糖尿病 90d仍持续较高水平。p5 3、Fas、Bcl -2和Bax分别于糖尿病 4 5d、4 5d、4 5d和 30d开始升高 ,并于糖尿病 6 0d、75d、75d和 6 0d达高峰。结论 糖尿病大鼠心肌细胞凋亡增加 ,凋亡相关蛋白的表达水平也增加。 Objective To study the cardiomyocytes apoptosis and expression of apoptosis-related proteins in diabetic(DM) rats. Methods Diabetic model was established by intraperitoneal injection with STZ, and apoptotic index, p53, Fas, Bax and Bcl-2 expression in myocardium were analyzed semiquantitatively by TUNEL or immunohistochemical technique in health or diabetic rats in 30d, 45d, 60d,75d and 90d, respectively. Results A few of apoptotic cells were seen in health rats, and both the immunoreactive positive and negative for p53,Fas, Bcl-2 and Bax were observed in health rats. 30d after DM, the apoptotic index significantly increased and remained high level till 90d after DM. The increase in p53, Fas Bax and Bcl-2 in the diabetic rats was evident in the 45d, 45d, 45d and 30d after DM and the peak level was seen in 60d, 75d 75d, or 60d, respectively. Conclusion The apoptosis is increased in diabetic myocardium and apoptosis-related proteins is also expression significantly.
