槐耳颗粒在乳腺癌综合治疗中的作用及其机制 被引量:25

摘要 目的 探讨金克 槐耳颗粒在乳腺癌综合治疗中的作用及其机制 ,并对其临床疗效进行观察。方法 采用四甲基偶氮唑蓝 (MTT)法测定药物的细胞毒性、耐药倍数和逆转倍数 ;应用荧光分光光度计测定细胞内ADM药物浓度 ;荧光定量PCR仪测定药物对细胞内MDR 1mRNA表达水平的影响。结果  (1) 0 .0 1mg ml剂量的槐耳颗粒能显著降低化疗药物ADM对人乳腺癌耐药细胞株MCF 7 A的IC50 (3.95 μg ml) ,与逆转前耐药细胞株MCF 7 A的IC50 (17.6 7μg ml)相比 ,差异有显著性 (P <0 .0 5 ) ,其逆转倍数为 4 .5倍 ,耐药倍数为 2 1.7倍。 (2 ) 0 .0 1mg ml槐耳颗粒能显著增加耐药细胞内ADM浓度 ,实验组与对照组相比 ,差异有显著性 (P <0 .0 1)。 (3)人乳腺癌敏感细胞株MCF 7 S无MDR 1mRNA表达 ,低毒剂量和非细胞毒性剂量的槐耳颗粒使MCF 7 A细胞的耐药基因MDR 1表达水平下调 ,与对照组相比差异有显著性 (P <0 .0 1)。 (4)临床观察发现 ,槐耳颗粒在乳腺癌综合治疗中对化疗有增敏作用。结论 金克 槐耳颗粒具有逆转MCF 7 A细胞耐药性的作用 ,逆转机制是使耐药基因MDR 1表达水平下调。槐耳颗粒在乳腺癌综合治疗中具有双效性 ,一是抗肿瘤活性 ,二可作为耐药逆转剂。 Objective To explore the function of the Jinke-huaierkeli plaster in the comprehensive therapy of breast cancer and the mechanism.Methods MTT method was used to detect the cytotoxicity of the drug,drug-resistant multiple and reversing multiple.Fluorescent spectrophotometer was used to measure the concentration of ADM in cells and fluorescent quantitative PCR method was used to detect the effect of the drug on the expressing level of MDR-1 mRNA in cells.Results (1) 0.01 mg/ml Huaierkeli plaster can significantly reduce the IC 50( 3.95 μg/ml) of ADM to the drug-resistant cell strain (MCF-7/A) of breast cancer,with significant difference as compared with the IC 50( 17.67 μg/ml) to the drug-resistant cell strain MCF-7/A without reversing(P<0.05).Its reversing multiple was 4.5 times and drug-resistant multiple was 21.7 times.(2) 0.01mg/ml Huaierkeli plaster could significantly increase the concentration of ADM in drug-resistant cells,with significant difference(P<0.01).(3) MDR-1 mRNA was not expressed in the sensitive cell strain MCF-7/S of human breast cancer.The Huaierkeli plaster at dosages of low toxicity or without toxicity lowered the expressing level of MDR-1 in MCR-7/S with significant difference as compared with the control(P<0.01).Conclusions This research indicate that Jinke-huaierkeli plaster can increase the sensitivity of breast cancer cells to chemotherapy drug.Its mechanism is that it can lower the expressing level of the drug-resistant gene MDR-1 and reverse the drug-risistance.At the same time the research preliminarily indicates that Jinke plaster has two effects,one is the anti-tumor activity and the other is reverse the drug-resistance.
