转染野生型p53基因对人肝癌细胞生长的抑制作用 被引量:10

摘要 目的 研究转染野生型 p5 3基因对人肝癌细胞的生长影响作用。 方法 将克隆有野生型 p5 3基因的 pUHD10 3质粒 ,通过Lipofectin转染肝癌细胞株BEL 74 0 2、BEL 74 0 4、HLE、HuH 7及SMMC 772 1,用四甲基偶氮唑 (MTT)法观察细胞的生长情况。结果 转染野生型p5 3基因 ,可使肝癌细胞BEL 74 0 4、HLE、HuH 7的生长明显受到抑制 ,第 4天生长抑制率分别是5 0 .0 %、6 9.0 %及 6 5 .1%。结论 转染野生型 p5 3基因能有效抑制肝癌细胞株BEL 74 0 4、HLE、HuH Objective To observe the growing effection in hepatocarcinoma cells of transfection with wild-type p53(wt-p53)gene.Methods The hepatocarcinoma cell line BEL-7402,BEL-7404,HLE ,HuH-7 and SMMC-7721 were transfected by the Plasmids pUHD10-3 recombined with wild-type P53 gene at lipofectin-mediated. The growing tests of the hepatocarcinoma cells were studied by monotetrazolium(MTT) analysis.Results The growth of the hepatocarcinoma cell line BEL-7404,HLE as well as HuH-7 were observably suppressed after transfection with the wt-p53 gene.Conclusion The wt-p53 gene effectively suppressed the growth of hepatocarcinoma cell line BEL-7404,HLE and HuH-7.
