摘要
目的分离、鉴定云南重楼ParispolyphyllaSmithvar .yunnanensis(Franch)HandMazz根茎中的甾体皂苷类化合物。方法采用乙醇提取、水饱和正丁醇萃取、硅胶柱色谱及高效液相色谱等方法进行分离 ,得到 5个化合物 ,通过FAB MS、1H NMR、13 C NMR、1H 1HCOSY、13 C 1HCOSY、HMBC、NOESY和TOCSY鉴定其化学结构。结果分离鉴定了 5种甾体皂苷 :薯蓣皂苷元 3 O {α L 鼠李吡喃糖基 (1→ 4 ) α L 鼠李吡喃糖基(1→ 4 ) [α L 鼠李吡喃糖基 (1→ 2 ) ]} β D 葡萄吡喃糖苷 (皂苷Pb ,Ⅰ )、纤细薯蓣皂苷 (Ⅱ )、皂苷Pa(Ⅲ )、薯蓣次苷A(Ⅳ )、pennogenindiglycoside(Ⅴ )。 结论化合物Ⅰ~Ⅴ均为已知甾体皂苷 ,纠正了已有文献中皂苷Pb(Ⅰ )核磁数据中的错误 ,提供了完整的NMR数据。
Aim To isolate steroidal saponins from the rhizomes of Paris polyphylla Smith var.yunnanensis(Franch)Hand Mazz and to identify their structures.Methods The compounds were separated by means of solvent extraction,chromatography on silica gel and HPLC,and their structures were elucidated on the basis of spectral analyses(FAB-MS, 1H-NMR, 13C-NMR, 1H- 1H COSY, 13C- 1H COSY,HMBC,NOESY and TOCSY).Results Five steroidal saponins were isolated from the rhizomes of Paris polyphylla Smith.They were elucidated as diosgenin 3-O-{α-L-rhamnopyranosyl(1→4)-α-L-rhamnopyranosyl(1→4)-[α-L-rhamnopyranosyl(1→2)]}-β-D-glucopyranoside(Ⅰ),gracillin(Ⅱ),saponin Pa(Ⅲ),prosapogenin A of dioscin(Ⅳ),pennogenin diglycoside(Ⅴ),respectively.Conclusion Compounds Ⅰ~Ⅴ are known as steroidal saponins.The extensive 2D NMR spectroscopies of compound Ⅰ,named saponin Pb,were studied and the NMR data were provided,as well as the correction with the previous incorrect assignment data.
