菘蓝的4种单体成分抗柯萨奇病毒作用的研究 被引量:4

摘要 目的 :了解菘蓝 4种有效单体 (6 ,7,8,9号 )抗柯萨奇病毒B组 3型 (CVB3 )的活性。方法 :通过观察细胞病变效应 (CPE) ,噻唑蓝 (MTT)比色法检测细胞活性 ,用半定量RT PCR检测不同药物浓度下病毒核酸含量 ,以确定有效单体抗CVB3 的作用。结果 :菘蓝的 4种单体成分对CVB3 生物合成均有抑制作用 ,且强于阳性对照药物病毒唑。表现在随着药物浓度的增加 ,CPE程度逐渐减弱 ,病毒核酸的含量逐渐下降。结论 :菘蓝的 4种单体能在体外通过抑制生物合成发挥抗CVB3 作用。 Objective: To investigate the antiviral activity of the four monomers of Isatis tinctoria L (the sixth, seventh, eighth, and ninth) on Coxsackievirus B 3 (CVB 3 ) in vitro. Methods: Anti-CVB 3 effects were investigated by observing cytopathic effect (CPE), by using MTT colorimetric assay for viable cell rate, and by detecting CVB 3 -RNA using semi-quantitative RT-PCR. Results: The four monomers of Isatis tinctoria L were more effective than the ribavirin in anti-biological synthesis of Isatis tinctoria L. As the dose of the drugs and the degree of CPE increased, the quantity of CVB 3 -RNA in culture media decreased and the viral inhibition rate increased. Conclusion: The four monomers of Isatis tinctoria L may inhibit CVB 3 biological synthesis but not inactivate the virus directly or block the adsorption to susceptible cells in vitro. Anti-CVB 3 effect of the four monomers was from best to worst as: the ninth monomer, the eighth monomer, the sixth monomer, and the seventh monomer.
