摘要 目的 研究人脑胶质瘤中细胞周期素E(cyclinE)表达与胶质瘤细胞恶性程度及细胞增殖活性的关系。方法 采用免疫组化方法检测 5 2例人脑胶质瘤和 8例正常脑组织标本中cyclinE和增殖细胞核抗原 (PCNA)的表达水平 ,观察并分析各病理等级中两者表达的阳性率和标记指数。结果 正常脑组织中无cyclinE或PCNA表达。随着人脑胶质瘤病理分级的增高 ,cyclinE的阳性率和平均标记指数均明显升高 ,在高、低度恶性度肿瘤之间也存在明显差异 (P <0 .0 5 )。双元相关性分析发现cyclinE与PCNA的平均标记指数呈明显正相关 (Pearson相关系数r =0 .5 76 ,P <0 .0 1)。 结论 人脑胶质瘤中cyclinE的表达与细胞增殖活性和病理学分级关系密切 ,很可能对肿瘤细胞的增殖和恶性转化产生重要的推动作用。 Objective To study the expression of cyclin E in human gliomas and its relationship with pathological grades and cellular proliferation activity.Methods The expression of cyclin E and proliferating cell nuclear antigen (PCNA) was detected by immunohistochemistry in 52 human brain gliomas and 8 normal brain tissues to observe their positive rates and the labeling indices in each WHO grade of human brain gliomas. Results Neither cyclin E nor PCNA was expressed in normal brain tissues. In human brain gliomas, positive rates and mean labeling indices for cyclin E increased accordingly with the advancing pathological grades. Significant difference was seen in labeling indices between low grade (grade Ⅰ or Ⅱ) and high grade (grade Ⅲ or Ⅳ)( P < 0.05 ). Positive correlation was obtained between the labeling indices for cyclin E and PCNA by bivariate correlation analysis (Pearson correlation coefficient r =0.576, P <0.01). Conclusion Expression of cyclin E is closely correlated with the cellular proliferation activity as well as pathological grades in human brain gliomas. It may be an important factor in promoting cell proliferation and transformation to malignant phenotype.
