重复电针预处理对兔脊髓缺血损伤后热休克蛋白90表达的影响 被引量:11

摘要 目的 :探讨重复电针刺激“足三里”穴对脊髓缺血性损伤保护作用的部分机理。方法 :1 6只成年雄性新西兰大白兔随机分成两组 (各组n =8) ,即对照组、电针预处理组。对照组每天静脉给予戊巴比妥钠 3 0mg/kg,连续 5d ;电针预处理组每天静脉给予戊巴比妥钠 3 0mg/kg,同时给予电针刺激双侧“足三里”穴 ,3 0min/d ,连续 5d。最后 1次预处理结束后 2 4h ,夹闭肾下腹主动脉2 0min ,制作兔脊髓缺血模型 ;并于 48h后处死动物取脊髓 (L5~ 7)制作标本 ,行免疫组织化学染色。结果 :再灌注 48h脊髓前角的HSP 90表达电针预处理组明显多于对照组 (P <0 .0 1 )。结论 :重复电针刺激“足三里”行预处理诱导缺血耐受的机理可能与增强HSP 90在脊髓前角的表达相关。 Objective: To investigate the influence of pretrea tment with repeated electroacupun cture(EA)of “Zusanli”(ST 36) on the expression of HSP 90 in spinal cord isch em ia-reperfusion injury rabbits. Methods: 16 male New Zealan d white rabbits (2.0 ~2.5 kg) were randomly and evenly assigned to control group that received sodiu m pentobarbitone 30 mg/kg (i.v.) everyday continuosly for 5 days, and EA group tha t received anesthesia and EA of “Zusanli”(ST 36) 30 min a day for 5 days. Twenty-four h ours after the last treatment, spinal cord ischemia was induced by an infrarenal a ortic occlusion for 20 min. 48 h after ischemia-reperfusion, the rabbits we re killed and heat shock protein (HSP) 90 expression of the spinal cord (L 5~L 7) tiss ue was assayed with immunohistochemical method. Results: No signific ant differences we re found between two groups in mean femoral arterial pressure (MAP), rectal temp erature, pH, partial pressure of oxygen (PaO 2), partial pressure of CO 2 (PaC O 2) and glucose content of the femoral arterial blood. HSP 90 expression in the ante r ior horn of the spinal cord in EA group were significantly higher than that in c ontrol group 48 h after ischemia-reperfusion (P<0.01). Conclusion: Repeatitive el ectroacupuncture pretreatments at “Zusanli” can induce upregulation of HSP 90 e xpression in the lumbar spinal cord after infrarenal aortic occlusion, which may contribute to the protection effect of EA on the ischemic injured spinal cord.
