抗条锈小滨麦易位系的鉴定 被引量:15

摘要 从八倍体小滨麦与普通小麦杂种后代中筛选到一个农艺性状优良的抗条锈小滨麦种质系山农 0 0 96 ,细胞学观察结果表明其根尖细胞染色体 2n =4 2 ,花粉母细胞减数分裂中期Ⅰ (PMCMⅠ )染色体构型为 2n =2 1Ⅱ ;它与小麦亲本的杂种F1 PMCMⅠ染色体构型平均值为 2n =19Ⅱ +1Ⅳ。RAPD分析表明 :在 10 0个十聚体核苷酸随机引物中 ,有 3个引物在山农 0 0 96中扩增出滨麦草的特异DNA带 ,它们分别记为S2 4 1 2 50 、S38750 、S4 2 6 40 ,初步确定山农 0 0 96是一个小滨麦易位系。以滨麦基因组DNA为探针的荧光原位杂交结果进一步表明山农 0 0 96是小麦与滨麦的小片段易位系。 A Tritileymus germplasm line, Shannong 0096, with useful agronomic characteristics and good resistance to stripe rust, was selected from the progeny of Octoploid Tritileymus and common wheat.The cytological results showed that the chromosome number of Shannong 0096 in root tip cell was 2n=42, its chromosome configuration at PMC MⅠ was 2n=21Ⅱ, and mean chromosome configuration of hybrids F 1 between it and wheat parent at PMC MⅠ was 2n=19Ⅱ+ 1Ⅳ. The random amplified polymorphic DNA (RAPD) analysis was made in the Shannong 0096.Among 100 random primers, 3 primers were revealed specific DNA bands of Leymus mollis designated as S24 1250 ,S38 750 ,S42 640 respectively.Therefore Shannong 0096 can be primarily identified as a Tritileymus translocation Line.The results of genome in situ hybridization(GISH) with the probe of Leymus mollis genome DNA, further proved that Shannong 0096 is a wheat-Leymus mollis translocation line with small fragment.
