摘要
研究了新试剂(对氯偶氮安替比林)与Ca2+的显色反应,建立了测定工业微生物胞液中微量Ca2+的新方法.在氢氧化钠介质中,对氯偶氮安替比林与Ca2+发生灵敏的显色反应生成1∶2的蓝色络合物.钙配合物最大吸收波长位于635nm,表观摩尔吸光系数为2.1×104L/(mol·cm).在25mL溶液中,0~20μgCa2+符合比尔定律.室温下,对氯偶氮安替比林与Ca2+显色反应立即完成,配合物的吸光度可以稳定24h以上.此外,该显色体系还具有良好的选择性,大多数常见金属离子、蛋白等活性成分均有较高的允许量,用于生物胞液中Ca2+的直接测定,结果令人满意.
A new reagent p-chloroantipyrylazo was studied as analytical reagent for spectrophotometric determination of Ca^(2+) in the cytolymph of industrial microorganis. In NaOH solution, p-chloroantipyrylazo can react with Ca^(2+) to form a 1:2 blue complex. The maximum absorption of this complex lie at 635 nm. Its apparent molar absorptivity is 2.1×10^(4) L/(mol·cm). Beer's law is obeyed for 0~20 g Ca^(2+)/25 mL. Under room temperature, the color reaction of Ca^(2+) with the reagent can complete instantly and the absorbance of the complex stable for 24 h at least.Moreover, the proposed method has good selectivity as well. Most metal ions and protein et al. can be tolerated in considerable amounts. It has been applied to determine calcium(II) in in the cytolymph of industrial microorganis with satisfactory results.
