摘要
目的 探讨谷氨酸对体外培养的大鼠皮层神经细胞的损伤作用。方法 在对体外新生大鼠(0~id)大脑皮层神经细胞进行原代培养的基础上,分组加入不同浓度的谷氨酸,作用10分钟,24小时后测定细胞死亡率、乳酸脱氢酶(LDH)活性及超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量。结果 培养大鼠皮层神经细胞在谷氨酸50μmol/L作用10分钟后,细胞死亡率和LDH活性明显增加,SOD活性显著降低,神经细胞MDA含量则明显增高。结论 谷氨酸可损伤培养大鼠皮层神经细胞,这种作用在某种程度上是由氧自由基介导的。
Objective To investigate the effect of glutamate on cultured neonatal rat cerebellar cortical neurons. Methods On the basis of primary in vitro culture of cortical neurons of neo-borne rat (0-1d), the neuron were exposed with glutamate for 10 minutes, and the lactate dehydrogenases (LDH), superoxide dismutase(SOD),malondialdehyde(MDA) in the media and the cell viability were assayed. Results It was found that the LDH level.content of MDA and the cell loss in the glutamate (50μmol) were markedly higher than those in the control,however ,the activities of SOD decreased. Conclusion The results indicate that the glutamate does have neurotoxicity ,which may be mainly directed by oxygen free radical.
