不同单位保种的艾氏腹水癌细胞核型及DNA含量的比较研究

Comparison Studies on the Karyological Characteristics and DNA Contents of Ehrlich Ascites Carcinoma Cells from 3 Institutes

目的 比较研究三个单位保种的艾氏腹水癌细胞（EAC）染色体数目及DNA含量。方法 取腹腔接种EAC 6 d的KM小鼠，腹腔注射秋水仙碱，取腹水，经低渗处理，固定后涂片，染色，显微镜下染色体计数；腹水传代的EAC，用70%乙醇固定，流式细胞仪测DNA含量。结果 山东省医学科学院药物研究所（山东）、武汉大学典型培养物保藏中心（武汉）和北京市肿瘤研究所（北京）保种的艾氏腹水癌细胞株，其染色体均数分别为28.31±7.78、29.16±12.25、23.22±10.21条，方差分析表明，除北京与山东外，武汉与北京、武汉与山东两两比较有极显著差异（P<0.01）。直方图分析显示主流染色体范围分别为25-27、22-24、22-24条。北京艾氏腹水癌细胞 DNA含量最多，山东艾氏腹水癌细胞DNA含量最少。结论 三个单位的艾氏腹水癌细胞株在某些方面已出现差异。

Objective To compare the chromosome number and DNA content of the Ehrlich ascites carcinoma(EAC) cell lines from 3 different institutes. Methods The KM mice were injected intraperitoneally with 0.2ml solution of EAC cell. After six days, the animals were injected intraperitoneally with 0.5ml solution of 0.025% colchicine. After 4 hours,the ascitic fluid was taken and 0.75mol hypotomic Kalium Chloride solution added. After fixation, we made a slide and examined the chromosome numbers of 100 cells under the microscope; EAC ascites tumor cells were fixed with 70% ethanol, DNA analysis by Flow Cytometry. Results The average chromosome numbers of the EAC cells were 28.31±7.78, 29.16±12.25,23.22±10.21 stored in Medicine Institute attached to Shandong Academy of Medical Sciences, the Type Culture Collection attached to Wuhan University and Beijing Cancer Institute ,respectively. The ANOVA indicated that chromosome numbers had significant defference both in Beijing and Shandong(P<0.01), as comparied with ones of Wuhan. The histogram showed that the main range of chromosome numbers varied between 25-27，22-24，22-24. Flow cytometry showed that DNA content of the cells from Beijing's was the most and that from Shandong's was the least. Conclusion These results testified that the EAC cells from 3 different institutes had changed to a certain extent.
