丹参对鼠脑缺血再灌注后白细胞介素-1β转化酶表达的影响 被引量:17

摘要 研究丹参在脑缺血再灌注后对白细胞介素 1β转化酶 (ICE)表达的影响及其神经保护作用。 方法 利用沙土鼠前脑缺血再灌注模型 ,应用RT PCR结合免疫组化的方法进行研究。结果 在脑缺血再灌注后 1~ 4d ,皮层及海马可见ICEmRNA表达。免疫组化结果显示脑缺血再灌注后脑组织ICE阳性细胞数显著增多 ,而丹参组ICE表达显著低于缺血对照组 ,但仍显著高于假手术组 (P <0 .0 1) ;丹参预防性干预组与丹参治疗性干预组相比 ,差异也有显著性意义 (P <0 .0 1)。结论 丹参可能通过调节ICE ,进而发挥治疗脑缺血的作用。 Objective To study the protective effect of radix salviae miltiorrhizae (RSM) through regulating the expression of interleukin 1β converting enzyme (ICE) in gerbil brain after global forebrain ischemia and reperfusion.Method On a model of global forebrain ischemia and reperfusion,RT PCR and immunohistochemical technique were used.Results Increased expression of ICE mRNA was induced in cortex and CA1 during 1~4 days after ischemia and reperfusion.Using immunohistochemical technique,it was observed that the number of ICE positive cells in brain increased significantly after global forebrain ischemia and reperfusion; although ICE expression in RSM group was lower than that in control group,it was still higher than that in sham operation group(P<0.01).Significant difference was also observed between the RSM prevented group and the RSM treated group(P<0.01).Conclusion RSM might play a neuroprotective role through regulating ICE expression during cerebral ischemia and reperfusion.
