摘要
目的 对西双版纳小耳猪的群体遗传结构进行RFLP分析。方法 采用ECL核酸直接标记和检测系统 ,以HRP标记浕 -珠蛋白 - 3’HVR多基因座小卫星探针 ,对西双版纳小耳猪的HinfⅠ或HaeⅢ酶切片段进行Southern杂交 ,以化学发光法进行检测 ,获得了清晰可辨的、具有高度多态性的DNA指纹图谱。结果 2kb以上的谱带数在 6~ 1 5条之间 ,HinfⅠ酶切产生的图带数低于HaeⅢ。JB亚系内 80 5、 80 7家系和JS亚系内 1 1 1、 1 2 1、1 2 1、 1 51家系不同个体间的相似系数分别为 0 94± 0 0 9、 0 85± 0 0 8、 0 84± 1 7、 0 78±1 6、 0 83± 0 42、 0 87± 0 0 7,显著高于各家系间的相似系数 ,JB亚系内的 80 5家系和JS亚系内的 1 51家系不同个体间相似系数较大 ,分别为 0 94和 0 87。结论 西双版纳小耳猪近交程度较高 ,个体间差异较小 。
Objective RFLP analysis was conducted to the genetic structure of Banna minipig inbred lines Methods Clearly and highly polymorphic DNA fingerprints from Banna miniature pig were obtained by southern hybridization with a nonisotopically HRP(HorseRadish Peroxidase) labelled multiloci minisatellite probe ɑ-globin 3'HVR,and detected by enhanced luminescence Results The fragment number of over 2kb ranged from 6 to 15,an more fragments resulted from the digestion of HinfⅠthan those of HaeⅢ The similarity coefficients of various individuals from family 805,807 in substrain JB and family111,121,131,151 in substrain JS were 0 94±0 09,0 85±0 08,0 84±1 7,0 78±1 6,0 83±0 42,0 87±0 07,which was higher than that between families Furtherly,simi_larity coefficients of family 805 in substrain JB and family 151 in substrain JS were 0 94 and 0 87 respectively,which were higher than others Conclusion The two families have the higher inbreeding degree,with the smaller genetic variation among individuals and the better homogeneity
