摘要
目的观察肺炎对幼鼠心肌细胞内游离钙离子浓度[Ca^(2+)]_i的影响,探讨肺炎时心肌功能障碍发生的机理.方法采用气管内接种的方法,建立幼鼠金黄色葡萄球菌肺炎模型,分离心肌细胞,运用钙离子荧光指示剂Fura-2/AM,检测心肌细胞内[Ca^(2+)]浓度.结果对照组24h(116.3±15.8)72h(120.5±12.6)和120h(118.2±13.5)相比,肺炎组接种后24h(138.7±13.6)(P<0.05)、72h(161.0±15.2)(P<0.01)和120h(173.6±11.2)(P<0.01)心肌细胞内游离钙含量升高.结论肺炎可引起幼鼠心肌细胞内钙超载,心肌细胞内钙稳态的变化可能是肺炎影响心肌功能的重要病理生理基础之一.
Objective To elucidate the effects of pnuemonia on the myocardial cells free calcium and its mechanisms. Methods A peumonia mode has been made with staphylococcus. The myocardial cells was isolated by collagenase. the free calcium contents of myocardial cells were measured with fluorescein fura -2/AM at 24h(116.3±15.8), 72h(120.5±12.6), 120h(118.2±13.5) of inoculation. Results Compared to control group, the free calcium contents of myocardial cells increased at 24h(138.7±13.6) (p<0.05), 72h(161.9±15.2)(p<0.01),120h(173.6±11.2)(p<0.01) in the pneumonia group rats. Conculation The changes of free calcium contents of myocardial cells may be one of the important biochemical indicators for cardiae function alterations associated with pneumonia.
