摘要
目的:为了探索临床热灭活恶性骨肿瘤细胞的适宜温度。方法:应用MTT法测定SDH活性及肿瘤干细胞集落形成法对HOS-8603细胞株进行体外热灭活(42℃~50℃)实验研究。结果:45℃60min或50℃10min以上时,HOS-8036已基本失去细胞集落形成能力。SDH的活性随温度升高及时间延长而下降,与细胞集落形成能力有较好的相关性。结论:以上研究提示临床上无需采用煮沸方法,灭活温度在50℃左右即可将肿瘤细胞杀灭。
Purpose:To explore the optimal temperature suitable to devitalize the osteosarcomacell. Methods:MTT assay test was performed to survey SDH activity, and colony-forming test of tumor stem cells was put to use to exterminate in vitro the Hos-9603 cell strain, with hyperthermia ranging from 42℃ to 50℃ for 10 minutes. Results: SDH activity decreased with rising of temperatures, which was relevant to cell ability of forming colony. Conclusions:Tumor cell can be dentaliged clinically of temper-ature of 50℃ rather than being boiled.
