摘要
目的:研究胰岛素样生长因子Ⅰ(IGF-1)对大鼠皮肤创面愈合过程的影响。方法:以大鼠为动物模型,在伤后1d、4d、7d、11d、14d、和17d取材,利用免疫组织化学和图像分析技术,研究皮肤创伤愈合过程中创面内源性IGF-1、增殖细胞核内抗原(PCNA)及Ⅰ、Ⅲ型胶原的动态变化,以及与创面愈合相关因素成纤维细胞之间的关系。结果:在皮肤创伤愈合过程中,内源性IGF-1表达量的变化与成纤维细胞活动、PCNA表达及Ⅰ型胶原的变化基本相符,与Ⅲ型胶原的变化关系不密切。结论:IGF-1与成纤维细胞增殖、活跃程度密切相关,是影响创面愈合的重要因子之一。
Objective:To study the effect of Type I Insulin like growth factor ( IGF-1 ) in the process of wound healing in the back skin of rat. Methods:The wound models were made in the back skin of rat, then the specimens in the wound site were harvested after 1, 4, 7, 11, 14, 17days. Immunohistochemistry(IHC) and Image analysis was employed to detect the change of regularity endogenous IGF-1 , Type Ⅰ,Ⅲ collagen and Proliferating cell nuclear antigen (PCNA)in the process of wound healing. Research the relationship between them and their related factor of wound healing: fibroblast. Results:The changes of IGF-1 in wound mainly coincident to the changes of fibroblast, PCNA, but have not closely relation with the changes of TypeⅢ collagen in the process of wound healing in the skin of rats. Conclusion:IGF-1 is very important in the process of wound healing. IGF-1 have a closely relation with the proliferation of fibroblast. It is one of the main factors of adjusting the excretion of Type Ⅰcollagen in wound by fibroblast.
