Expression and Significance of Transforming Growth Factor β_1 mRNA in Colorectal Carcinoma Tissue and Cell Line

研究转化生长因子（ＴＧＦ）β_１ｍＲＮＡ在大肠癌癌组织、癌旁和癌远端组织及ＨＲ－８３４８细胞系中的表达，探讨其在大肠癌发生、发展中的作用。方法：取１４例大肠癌患者的癌组织、癌旁２ｃｍ和１０ｃｍ全层肠管及ＨＲ－８３４８细胞系，用Ｎｏｒｔｈｅｒｎ ｂｌｏｔ法检测ＴＧＦ－β_１ｍＲＮＡ的表达。结果：①１４例大肠癌癌组织、癌旁和癌远端组织及ＨＲ－８３４８细胞系中均有ＴＧＦ－β_１ｍＲＮＡ不同程度的表达，以癌远端组织表达最弱，１４例病例癌远端组织中ＴＧＦ－β_１个ｍＲＮＡ的表达均呈减弱趋势；②在癌远端组织中，Ｄｕｋｅｓ Ａ、Ｂ期的３例和Ｃ期的２例杂交信号明显弱于其他Ｃ、Ｄ期病例。结论： ＴＧＦ－β_１在大肠癌发生、发展中起重要作用，其ｍＲＮＡ表达与Ｄｕｋｅｓ分期相关，可望作为大肠癌恶性程度的指标。ＴＧＦ－β-１ｍＲＮＡ在癌远端组织中表达减弱的趋势是否可作为临床手术切缘的评估指标，有待进一步研究。

Background/Aims: To investigate the expression of transforming growth factor (TGF) β_1 mRNA in HR-8348 cell line and 3 different segments of colon tissue specimens from 14 cases with colorectal carcinoma, and its role in the development and growth of colorectal carcinoma. Methods: The speci- mens were taken from cancer tissue, paracancerous tissue (2cm away from the tumor), and distal area (10cm away from the tumor) from 14 cases with colorectal carcinoma. Northern blot assay was used to detect the expression of TGF-β_1 mRNA in these tissues and the cell line. Results: 1 TGF-β_1 mRNA expressed in varying degree in HR-8348 cell line and the three groups of tissue specimens from those cases of colorectal carcinoma. The expression was the weakest in the distal area, and in the same case, it showed a declining tendency toward the distal area; 2 The hybridized signals in 3 cases with Dukes A and B, and 2 cases with Dukes C were significantly weaker than other cases with Dukes C and D. Conclusions: TGF-β_1 plays a key role in the oncogensis of colorectal carcinoma, and the expression of TGF-β_1 mRNA was related to Dukes staging. It is anticipated to be a marker of malignant degree of colorectal carcinoma. Furthermore, whether the decreased expression of TGF-β_1 mRNA in the distal area might be able to serve as a marker for evaluating the margin of excision in clinical practice remains to be investigated.
