摘要
目的 建立抗感清胶囊中连翘苷含量的高效液相色谱测定法。方法 样品用80％乙醇提取后,以D101大孔吸附树脂柱分离杂质,洗脱液浓缩后测定。以Hypersil ODS柱(4.0mm×250mm,5μm)分离,乙腈-0.1％磷酸溶液(22:78)为流动相,检测波长277nm。结果 该样品处理条件合理,色谱峰分离完全,杂质不干扰测定,方法稳定可行,平均回收率为98.9％,RSD=2.02％(n=5)。结论 RP-HPLC法测定抗感清胶囊中连翘苷含量,方法灵敏、准确,可用于该制剂的质量控制。
OBJECTIVE To establish a RP - HPLC method for the determination of the pillyrin in kangganqing capsules. METHODS Samples were separated in a D-101 macroporous resin column after beiny extracted by 80% alcohol solution, and then the elute solution was concentrated and determined by a Hypersil ODS column (4.0 mm×250 mm, 5μm). The mobile phase of consisted acetonitrile-0.1 % phosphate buffer (22 -78), and phillyrin was detected at UV 277 nm. RESULTS The method of sample extraction was reasonable, and the chromatography of phillyrin was symmetry and was separated well from other substances. The average recovery was 98.9% , and within-day RSD was 2.02 % (n = 5). CONCLUSIONS The sensitive and reliable newly-developed RP-HPLC method is suitable for the determination of the phillyrin in the kangganqing capsules.
