摘要
9—去氢海柯皂甙元在荧光薄层上呈现荧光熄灭效应。应用这一效应,研究了9—去氢海柯皂甙元的荧光薄层扫描测定法。番麻皂素在硅胶GF_(204)薄层板上,用氯仿—丙酮(9:1)展开。分离的9—去氢海柯皂甙元斑点,用日立MPF—4型荧光分光光度计及其薄层色谱附件扫描。扫描条件:λex=254nm,λem=530nm,反射式、直线扫描。标准曲线浓度为1~4微克。以测量峰面积定量。
Fluorescent TLC densitometry was employed in the determination of g-dehydrohecogenin in the sapogenin of Americaa aloe(Agave americana)on the basis of the fact that 9-dehydrohecogenin displays fluorescent quenching effect on silica gel GF2-54 plates under uv-light. American aloe sapogenin was developed on silical gel GF254 plate with chloroform-acetone(9:1). The separated 9-dehydrohecogenin was then scanned(reflection mode, linear scanning)on a Hitachi MPF-4 fluorescent spectrophotometer combined with TLC accessory at λex=254nm and λem=530nm. The concentration of standard curve was in the range of 1-4 μg. Each sample was quantitated by measuring the area of its peak
