重组大肠杆菌生产hEGF发酵条件的研究

摘要 在摇瓶条件下 ,研究了发酵条件对重组大肠杆菌 JLU菌株 h EGF表达的影响 .研究表明 ,最适的发酵条件是温度在 32℃、p H6 .6~ 7.3、初始葡萄浓度 5g/ L、装液量 30 m L/ 2 50 m L三角瓶 ,并发现在发酵培养基中用 0 .5g/ L甘油代替 5g/ L葡萄糖作为发酵碳源会进一步提高 h EGF的表达水平 ,提高幅度可达 15.5% The effects of fermentation conditions on hEGF expression in recombinant \%E.coli\% strain JLU were examined in shake flask scale. It was found that initial glucose concentration and temperature were two key factors which affected hEGF expression. Maximum hEGF expression was observed when initial glucose was 5.0 g/L and the temperature was 32℃. Besides, high hEGF productivity was obtained when initial pH was adjusted between 6.6 and 7.3, and medium volume was 30 mL in 250 mL flask. The hEGF productivity could be further raised by 15.5% when 5 g/L glucose was replaced by 0.5 g/L glycerol as the carbon source in the fermentation medium.
