摘要
目的:探讨四物汤对环磷酰胺所致贫血小鼠肾促红细胞生成素基因表达的影响,从分子生物学水平为四物汤补血作用提供理论依据。方法:用环磷酰胺腹腔注射建立小鼠贫血模型,同时给小鼠灌服四物汤,采用Trizol试剂提取肾组织总RNA,反转录-聚合酶链反应(RT-PCR)方法检测肾组织促红细胞生成素mRNA表达情况(β-actin为内对照标准)。结果:四物汤组小鼠肾脏EPOmRNA(2.589±0.426)表达显著高于对照组(2.438±0.233)和环磷酰胺组(2.396±0.354),差异均有显著性意义(t=2.849,2.838,P均<0.05)。结论:四物汤能抑制环磷酰胺所致的小鼠贫血,其作用机制可能是通过促进肾组织EPOmRNA的表达,使肾脏合成促红细胞生成素增多,从而促进红细胞生长。
AIM:To investigate the effect of siwu tang on erythropoietin(EPO) gene expression in the kidney of mice with anemia induced by cyclophosphamide,and provide theoretical basis for the function of nourishing the blood of siwu tang on molecular biological level. METHODS:Anemia models were induced with the injection of cyclophosphamide in abdominal cavity of mice,and at the same time,the mice received siwu tang.Total RNA(ribonuclunc acid) was extracted with Trizol reagent.The EPO mRNA expression in kidney was measured by RT-PCR (β-actin gene as a control). RESULTS:The EPO mRNA expression levels of siwu tang group(2.589±0.426) was significantly higher than that of the control group(2.438±0.233) and cyclophosphamide group(2.396±0.354),and there was significant difference(t = 2.849,2.838,P< 0.05). CONCLUSION:The siwu tang has a good inhibitory effect on anemia induced by cyclophosphamide.The mechanism of siwu tang is to increase EPO production by promoting the EPO mRNA expression in kidney,so as to stimulate the growth of erythrocyte.
