摘要
目的 :探讨通脉宁胶囊 (TMNJN )对家兔颈内动脉内皮球囊损伤后血管狭窄模型血液流变学及相关活性因子的影响。方法 :在无菌条件下分离出颈内和颈外动脉 ,由颈外动脉向颈内动脉插入气囊 ,在 2 pa恒定压力下 ,反复抽拉3次 ,造成经腔内皮损伤和血管狭窄。连续给药 2 1天后 ,颈总动脉取血 ,分别测定各实验组家兔全血黏度、血浆黏度、血球压积、红细胞聚集指数、聚集面积、红细胞变形指数、变形面积及血浆血栓素 (TXB2 )、6-酮 (6-keto -PGF1α)含量的变化。结果 :同假手术组相比 ,模型组家兔在造型 2 1天后全血黏度、红细胞压积、红细胞聚集指数、血浆TXB2 、TXB2 / 6-keto -PGF1α含量及比值明显升高 ,6-keto -PGF1α、红细胞变形指数明显降低。同模型组相比 ,造型家兔灌服通脉宁胶囊后 ,大、中剂量组全血黏度、红细胞压积、红细胞聚集指数、TXB2 、TXB2 / 6-keto -PGF1α明显降低 ,6-keto -PGF1α、红细胞变形指数明显升高。结论 :通脉宁胶囊具有较强的活血化瘀作用 ,为通脉宁胶囊能治疗血管内皮球囊损伤引起的血管狭窄提供了充实的药理学基础。
Objective:To explore effects of Tongmaining Capsules on haemorheology and related active factors after internal carotid artery balloon injury in rabbits models of vascular stenosis.Methods:With asepsis operation,internal and external carotid artery was separated,then the balloon was only inserted from external carotid artery to internal carotid artery,and draw back and forth repeatedly for three times in constant pressure of 2 Pa,to result in carotid artery endodermis injury and vascular stenosis.After the doses were given successively for 21 days,the blood were taken from the common carotid artery,and whole blood viscosity,plasma viscosity,corpuscular volume,erythrocyte aggregation index,aggregation area,erythrocyte deformation index,deformation area,content of TXB 2 and 6-keto-PGF 1α in plasma were measured.Results:Compared with the sham operation group,whole blood viscosity,corpuscular volume,erythrocyte aggregation index,TXB 2 in plasma,and content and ratio of TXB 2 and 6-keto-PGF 1α in rabbits of model group increased significantly,but 6-keto-PGF 1α and erythrocyte deformation index decreased significantly.Compared with model group,in Tongmaining Capsules large and middle dose group, whole blood viscosity,corpuscular volume,erythrocyte aggregation index,TXB 2 in plasma,and content and ratio of TXB 2 and 6-keto-PGF 1α decreased significantly,and 6-keto-PGF 1α and erythrocyte deformation index increased significantly.Conclusion:Tongmaining Capsules can promote blood circulation to dispel blood stasis strongly,thus this study provide substantial pharmacological basis about therapy of vascular endodermis injury induced vascular stenosis.
