摘要
目的 观察芳维A酸乙酯 (arotinoidethylester ,AE)在HL 60细胞分化成熟过程中对其CD11b、CD15表达的影响 ,初步探讨AE治疗脓疱型银屑病的可能机制。方法 HL 60细胞以二甲基亚砜 (dimethylsulfoxide ,DMSO)、全反式维A酸 (all trans retinoicacid ,ATRA)、AE诱导培养 ,5d后用流式细胞仪检测HL 60细胞表面CD11b和CD15的表达。结果 HL 60细胞经DMSO、ATRA、AE诱导后CD11b和CD15荧光标记率均比诱导前提高 ;ATRA组HL 60细胞CD11b和CD15荧光强度显著低于DMSO组 ,AE组HL 60细胞CD11b和CD15荧光强度显著低于ATRA组。结论 AE可明显抑制HL 60细胞表达CD11b、CD15 ;AE可能通过抑制参与炎症反应的中性粒细胞表达CD11b、CD15而降低中性粒细胞参与炎症反应的过程。
Objective To observe the effects of arotinoid ethylester (AE) on the expressions of CD11b and CD15 during the differentiation process of HL-60 cells and to investigate the possible mechanisms for the treatment of pustular psoriasis (PP) with AE. Methods The expressions of CD11b and CD15 on differentiated HL-60 cells, cultured for 5 d after induction with dimethylsulfoxide (DMSO) at a final concentration of 1.3% (DMSO group), all-trans-retinoic acid (ATRA) at 1×10 -6 mmol/L (ATRA group), and AE at 1×10 -8 mmol/L (AE group) respectively, were examined by flow cytometry. Results The expressions of CD11b and CD15 on HL-60 cells were enhanced after induction with DMSO, ATRA, and AE. The expressions of CD11b and CD15 on HL-60 cells in both ATRA group and AE group were lower than those in DMSO group. The expressions of CD11b and CD15 on HL-60 cells in AE group were the lowest among the above-mentioned three groups. Conclusion AE can suppress the expressions of CD11b and CD15 on HL-60 cells during the differentiation process. AE may inhibit the inflammation of PP by affecting the expressions of CD11b and CD15 on polymorphonuclear leukocytes.
