摘要
目的 :探讨候选抑癌基因Caveolin 1在胃癌中的表达及其与胃癌生物学行为的关系。方法 :应用免疫组织化学SABC法对 43例胃癌存档蜡块组织及 15例胃炎活检标本进行Caveolin 1蛋白检测 ,采用 χ2 检验进行统计学分析。结果 :Caveolin 1在胃癌的阳性表达率为 3 0 2 % ( 13 / 43 ) ,明显低于胃炎组 10 0 % ( 15 / 15 ) ,P <0 0 1;其中高分化腺癌、中分化腺癌、低分化腺癌中的阳性率分别为 6/ 6、3 / 5、17 4% ( 4 / 2 3 ) ,而印戒细胞癌及黏液腺癌中无阳性表达 ,五者之间差异有统计学意义 ,P <0 0 1;浸润至浆膜或超过浆膜组的Caveolin 1表达阳性率13 8% ( 4 / 2 9)明显低于未达浆膜组 64 3 %( 9/ 14 ) ,两组之间差异有统计学意义 ,P <0 0 1;淋巴结转移组Caveolin 1阳性率 7 7%( 1/ 13 )明显低于无淋巴结转移组 40 0 %( 12 / 3 0 ) ,两组之间差异有统计学意义 ,P <0 0 5。结论 :候选抑癌基因Caveolin 1的表达与胃癌细胞的分化、浸润和转移密切相关 。
OBJECTIVE:To study the expression of Caveolin-1 in gastric carcionma cells, and the correlation between expression of Caveolin-1 and clinicopathlogical profiles including tumor differentiation,invasion,and lymph node metastasis. METHODS: The expression of Caveolin-1 was tested by the SABC immunohistochemical method in 43 cases of resected gastric carcinomas tissues and 15 cases of gastritis tissues,and the expressive level of Caveolin-1 was detected by Western blot in 5 cases of gastric carcinomas, noncancerous tissues and 2 cases of fetal stomach disease tissues.RESULTS:The positive rate of Caveolin-1 in gastric carcinomas was significantly lower than those of gastritis and noncancerous tissues and fetal stomach disease tissues. The percentages of positive cases for Caveolin-1 in gastritis and gastric carcinomas were 100% ( 15/15) and 30.2% (13/43) respectively,P<0.01. There was lower expression in low differentiation cancer cells than that in well differentiation.The deeper tumor invasion, the lower Caveolin-1 expression. Cancer with lymph node metastasis had less Caveolin-1 protein than that without lymph node metastasis.CONCLUSIONS:Expression of Caveolin-1 is associated with the differentiation and metastasis of gastric carcinomas which might be a valuable marker for the biological classifaction,invasion and lymph node metastasis of gastric carcionma cells.
