摘要 目的 :探讨葡萄胎与正常胎盘绒毛基因表达谱 ,研究葡萄胎滋养层细胞增生的机制。方法 :采用cDNA芯片技术 ,用Cy3 dUTP标记正常胎盘绒毛mRNA ,用Cy5 dUTP标记葡萄胎组织mRNA ,制备探针进行芯片杂交 ,随机抽取 3条基因作RT PCR验证。结果 :从 40 0 4个有效基因位点中筛选出 10 6个差异表达基因 ,占 2 65 % ,其中上调基因表达共 5 7条 ,占1 42 % ;下调基因共 49条 ,占1 2 2 %。获得了葡萄胎与正常胎盘绒毛基因表达的信息。结论 :葡萄胎滋养层细胞增生与多基因激活具有密切的联系 。 OBJECTIVE:To explore gene expression profiles of complete hydatidiform mole to understand mechanisms of trophoblastic cell hyperplasia from gene level. METHODS:Total RNA was isolated from tissue samples of complete hydatidiform mole and normal early placental villi. Fluorescent probes were prepared from RNA labeled with cy3-dUTP for the normal early placental villi groups and with cy5-dUTP for the complete hydatidiform mole groups through reverse-transcriptation. The intensity of each spot on the microarray and ratios of Cy5 and Cy3 were analyzed, and finally the differentially expressed genes were selected by GenePix Pro 3.0 software.Three differential expression genes were selected randomly and analyzed by β-actin semi-quantitative RT-PCR. RESULTS:Of the 4 004 human genes analyzed by microarray, 106 genes (holding 2.65% of the investigated genes) were revealed differential expression in complete hydatidiform mole groups compared with the normal early placental villi groups, of which 57 genes were up-regulated (holding 1.42% of the investigated genes) and 49 genes were down-regulated (holding 1.22% of the investigated genes), which supplied abundant information about differential expression genes in trophoblastic cell hyperplasia.CONCLUSIONS:The present study suggests hyperplasia of trophoblastic cell in complete hydatidiform mole is highly correlated with up-regulation and activation of a number of genes,and the complexity of trophoblastic cell to adjust the gene expression is revealed as a whole by the gene chip technology.
