摘要
目的探讨缺氧诱导因子1α(HIF1α)在大肠腺癌组织中的表达及其与细胞增殖、血管形成的关系。方法应用免疫组织化学方法检测20例正常大肠组织、60例大肠腺癌组织中HIF1α、增殖细胞核抗原(PCNA)蛋白表达及微血管密度(MVD),并分析HIF1α表达与PCNA、MVD的相关性。结果HIF1α蛋白在大肠腺癌中的阳性表达率为73.3%(44/60),其阳性表达与大肠腺癌淋巴结转移、Dukes分期有关,其阳性表达率淋巴结转移组(89.3%,25/28)显著高于无淋巴结转移组(59.4%,19/32)(P<0.05),并随肿瘤分期的升高而增高。HIF1α表达与PCNA、MVD呈正相关(γ=0.647,P<0.01和γ=0.655,P<0.01)。结论HIF1α在大肠腺癌组织中表达水平明显上调,并与肿瘤细胞增殖、血管形成密切相关,提示它可能在大肠癌的发生发展过程中起重要作用。
Objective To identify the expression of HIF-1α and its relationship with angiogenesis and cell proliferation in colorectal adenocarcinoma.Methods Immunohistochemical technique was used to detect the expression of HIF-1α、PCNA and MVD in 60 cases of colorectal adenocarcinoma and 20 cases of normal colorectal mucosa.The relationship between the HIF-1α expression and PCND、MVD was evaluated.Results Of 60 cases of colorectal adenocarcinoma,44 cases were positive for HIF-1α(73.3%).The expression of HIF-1α was associated with the Dukes stage and lymph nodes metastases.The expression of HIF-1α was positively correlated with the PCNA and MVD(γ=0.647,P<0.01 and γ=0.655,P<0.01).Conclusion The up-regulation of HIF-1α expression is closely related with angiogenesis and cell proliferation in colorectal adenocarcinoma.HIF-1α might play an important role in the carcinogenesis and development of colorectal adenocarcinoma.
