乳酸-羟基乙酸共聚物微球中蜂毒包封率的测定 被引量:2

摘要 目的 建立蜂毒乳酸 羟基乙酸共聚物微球包封率的测定方法。方法 用二甲基亚砜 (DMSO)和含有十二烷基硫酸钠的氢氧化钠溶液的混合溶剂将微球中溶解性相差较大的基质和药物同时溶解 ,形成均匀的一相 ,再用Lowry法测定其中多肽的含量 ,并将测定结果与已经过方法学验证的二喹啉甲酸法进行了比较。结果 蜂毒在 10 0 5~ 10 0 5 μg内Lowry法测定的结果线性关系良好 ,相关系数为 0 9996 ,方法回收率平均为 10 0 7% ,RSD为 1 1% ,测定结果与二喹啉甲酸法无显著性差异 (P <0 0 5 )。结论 此方法稳定、可靠 。 OBJECTIVE: To establish a method for the determination of the encapsulation efficiency of bee venom loaded PLGA microspheres. METHODS: The proper combination of water-miscible dimethyl sulfoxide and NaOH solution containing SDS made it possible to solubilize both the hydrophobic matrix of microspheres and the hydrophilic bee venom in a single phase. The actual content of bee venom in the final solution was determined by Lowry method. RESULTS: The linear relationship was obtained over the range of 10.05 ∼ 100.5 μg (r = 0.999 6). The mean recovery of bee venom PLGA microspheres was 100.7% with RSD of 1.1%. Compared with validated BCA method, the results had no significant difference(P > 0.05). CONCLUSION: The method was proved to be stable and reliable to determine the encapsulation efficiency of biodegradable PLGA microspheres.
