TSP-1和VEGF在肝细胞癌新生血管生成中的作用 被引量:19

Role of thrombospondin-1 and vascular endothelial growth factor in neovascularization of hepatocellular carcinoma

摘要 目的 研究血小板反应蛋白 1(TSP 1)和血管内皮生长因子(VEGF)的平衡在肝细胞 癌(HCC)新生血管生成中的作用。方法 采用免疫组织化学SABC法检测37例HCC及其癌旁肝 组织和6例正常肝组织中TSP 1、VEGF和CD34的表达情况,研究TSP 1和VEGF的表达与HCC 的临床病理资料及新生血管生成的关系。结果 37例HCC中TSP 1阳性率为62.2%(23/37),低 于相应的癌旁肝组织和正常肝组织(P<0.05)。HCC中VEGF的阳性率为97.3%(36/37),高于相 应的癌旁肝组织和正常肝组织(P<0.05)。有镜下静脉浸润的HCC组织中TSP 1的表达低于无镜 下静脉浸润者,而VEGF的表达则高于无镜下静脉浸润者(P<0.05)。等级相关分析显示,HCC中 VEGF的表达与CD34的表达呈正相关关系,(rs=0.333,P<0.05),而TSP 1的表达与CD34的表达 则呈负相关关系,相关系数(rs=-0.393,P<0.05)。结论 TSP 1低表达与VEGF过表达之间的 不平衡性是决定HCC组织中新生血管生成的关键因素之一。 Objective To study the role of the balance between thrombospondin-1 (TSP-1) and vascular endothelial growth factor (VEGF) in the neovascularization of hepatocellular carcinoma (HCC). Methods The expression of TSP-1, VEGF and CD34 in 37 samples o HCC and their corresponding paraneoplastic tissues and 6 samples of normal liver tissue were determined with immunohistochemistry. The relation of the expression of TSP-1 and VEGF to clinicopathological parameters and neovascularization was also analyzed. Results The rate of TSP-1 expression was lower but that of VEGF expression higher in HCC than in paraneoplastic tissue and normal liver tissue (P<0.05). The rate of TSP-1 expression was lower but that of VEGF expression higher in HCC with microscopic venous invasion than in that without the invasion (P<0.05). Searman correlation test showed that VEGF expression in HCC was positively but TSP-1 expression in HCC negatively correlated to MVD (r s=0.333, P<0.05 and r s=-0.393,P<0.05). Conclusions Umbalance between the down-expression of TSP-1 and overexpression of VEGF in HCC is one of the key factors for neovascularization in HCC.
