白血病细胞和正常细胞生成树突状细胞的比较研究 被引量:2

摘要 目的 体外诱导髓系白血病原代细胞分化生成树突状细胞 (dendritic cell,DC) ,并与正常的 DC比较。方法 分离初诊 2 4例急性髓细胞性白血病和 10例慢性髓细胞性白血病患者的骨髓单个核细胞以及 5例正常人外周血的单个核细胞 ,用 rh GM- CSF 10 0 0 U/ m1、rh IL- 4 5 0 0 U/ m l和 TNF- α5 0 U/ m l联合培养 10天 ;形态学(Wright染色、倒置显微镜、透射电镜 )、免疫学 (CD80、CD86、CD83、CD1a、HL A- DR)鉴定 ,RT- PCR检测白血病克隆以及混合淋巴细胞反应 (ML R)检测抗原递呈功能。结果 细胞因子诱导后 ,正常细胞和白血病细胞均出现典型形态的树突状细胞 ,CD80、CD86、CD83、CD1a的表达明显上调 (正常和 CML- DC的 HL A- DR也明显上调 ) ,急性和慢性髓细胞性白血病以及正常细胞来源的树突状细胞具有不同程度的刺激异基因 T淋巴细胞增殖的能力。结论 无论是急性和慢性白血病细胞均能诱导分化成 DC,并且特征完全相似 ;白血病与正常骨髓的 DC在形态和免疫学表达方面相似 ,但功能较弱。
