摘要
应用ISSR分子标记方法对采自广西和广东的 7个罗汉果 (Siraitiagrosvenorii)野生居群共 130个个体进行了遗传多样性分析。 15个ISSR引物共扩增到了 111个位点 ,其中 91个是多态性位点 ,占 82 .0 %。Nei′s基因多样性指数 (He)为 0 .2 4 8,Shannon信息多样性指数 (I)为 0 .35 4。罗汉果不同居群的遗传多样性水平差异较大 ,居群多态位点百分率在 2 8.2 % - 5 5 .6 %之间 ,Nei′s基因多样性指数为 0 .0 80 - 0 .2 0 9,Shannon信息多样性指数为 0 .12 3- 0 .310。永福居群 (YF)和金秀居群 (JX)的遗传多样性水平较高 ,其周边居群的遗传多样性水平逐渐降低 ,居群间产生了较大的遗传分化 (Gst=0 .5 6 9)。居群间的遗传距离与地理距离相关性不明显 (r =0 .36 9,P =0 .115 )。UPGMA聚类图中 。
Siraitia grosvenorii, a liana species of the Cucurbitaceae endemic to southern China, is an important economic plant. The genetic diversity of 130 individuals from seven Siraitia grosvenorii populations from Guangxi and Guangdong provinces was estimated using inter-simple sequence repeat (ISSR) markers. The fifteen primers employed produced a total of 111 scorable amplified fragments, of which 91 (82.0%) are polymorphic. Genetic diversity analysis showed that Nei′s gene diversity (H e) was 0.248 and Shannon′s genetic diversity index (I) was 0.354. Genetic differentiation mainly occurred among populations (G st=0.569). The correlation between genetic distance and geographic distance among the populations was not significant (r = 0.369, P=0.115). UPGMA cluster analysis showed that the 130 individuals were clustered into seven clusters corresponding to the seven populations.
