肝移植患者血清白细胞介素-6、细胞间粘附分子-1和P-选择素高水平表达分析 被引量:2

摘要 目的 :研究白细胞介素 - 6、肿瘤坏死因子 -α及细胞间粘附分子 - 1和P -选择素在稳定存活的肝移植受者血清中的表达程度。方法 :ELISA法检测 2 2例原位肝移植患者和 12例正常人血清白细胞介素 - 6、肿瘤坏死因子 -α及细胞间粘附分子 - 1和P -选择素表达情况。流式细胞术分析外周血T细胞表型。同时对近期 6例临床原位肝移植病例进行动态观察。结果 :肝移植病人外周血CD3+ 、CD4 + 、CD8+ T细胞百分比以及CD4 + /CD8+ 比值与正常人组间比较无明显差异 (P >0 . 0 5 ) ,但肝移植组CD3+ CD2 5 + T细胞百分比明显高于正常人组 (P <0 . 0 5 )。白细胞介素 - 6、细胞间粘附分子 - 1和P -选择素水平肝移植组显著高于正常人组 (P <0 0 5 )。近期 6例临床原位肝移植病例血清白细胞介素 - 6、肿瘤坏死因子 -α及细胞间粘附分子 - 1和P -选择素水平的动态观察 ,未发现规律性变化。结论 :细胞间粘附分子 - 1和P -选择素参与了肝移植后效应T细胞的活化过程 ,肿瘤坏死因子 -α和白细胞介素 - 6则可能分别介导了对移植物的免疫损伤和修复。目的 :研究白细胞介素 - 6、肿瘤坏死因子 -α及细胞间粘附分子 - 1和P -选择素在稳定存活的肝移植受者血清中的表达程度。方法 :ELISA法检测 2 2例原位肝移植患者和 12例正常人血? AIM: To study the profile of serum IL-6, I CA M-1 and P-selectin in stable survivors with clinical liver transplantation (LTx) . METHODS: Flow cytometric analysis was used to determine the phen otype of T cell subsets in peripheral blood mononuclear cells (PBMCs) from sta ble survivors with liver transplantation (n=22), and healthy volunteers (n =12). Serum levels of the pro-inflammatory cytokines, tumor necrosis factor (T N F-α) and interleukin (IL-6), intercellular adhesion molecules (ICAM-1) and P-se lectin in stable survivors with liver transplantation and healthy volunteers wer e assessed by enzyme-linked immunoabsordent assay (ELISA). Recently performed 6 cases of liver transplantation were also dynamically observed in this study. RESULTS: Percentage of CD4+ T cells, CD8+ T cells and CD3 + T cells, as well as ratio of CD4 to CD8 were no difference between two groups (P>0.05). However, a significant higher percentage of CD3+CD25+ T cells was found in stable liver transplantation group as compared to healthy group ( P<0.05). Significantly increased concentrations of IL-6, ICAM-1 and P-selec tin were found in stable liver transplantation group as compared to healthy grou p (P<0.05). A high TNF-α level was detected in stable liver transplantatio n group while no significant difference was found as compared to healthy volunte ers group (P>0.05). There was not found no regular change of serum cytokine s (IL-6, TN F-α) and adhesion molecules (ICAM-1, P-selectin) in 6 liver transplanted patien ts during post-operation from day 1 to day 30, indicating that was associated wi th the different status of patients before or after transplantation. CONCLUSIONS: Our data suggesting that increased levels of ICAM-1 and P-selectin, appears to participate in the processing of immunoregulation to transplanted livers, whereas elevated concentrations of IL-6 appear to be involv ed in the repair of the injury induced by TNF-α in allo-transplanted livers.
