慢性粒细胞白血病树突状细胞诱导及其抗肿瘤功能的体外研究

摘要  目的 探讨人慢性粒细胞白血病(CGL)来源的树突状细胞(DC)的免疫功能。方法 应用重组人粒 巨噬细胞集落刺激因子(rhGM -CSF)联合重组人白介素 (rhIL) 4在体外诱导培养CGL细胞,获得DC。应用流式细胞仪(FACS)检测诱导前、后的细胞表型,观察其诱导的细胞毒T淋巴细胞 (CTL)的体外抗肿瘤效应。采用酶联免疫吸附试验(ELIS)测定DC培养及DC与外周血单个核细胞(PBMNC)共同培养的上清液中IL- 12及干扰素(IFN) γ的量。结果 联合rhGM- CSF及rhIL- 4可诱导CGL细胞分化为DC;CGL -DC体外激活的CTL对CGL细胞具有特异性细胞毒活性;分别从诱导DC培养上清液和DC+PBMNC培养的上清液中检测出一定量IL- 12及IFN γ。结论 CGL- DC具有抗原提呈细胞的免疫功能,可望用于CGL的免疫治疗。 Objective To investigate the immune function of dendritic cells derived from chronic granulocytic leukemia(CGL-DC). Methods After being treated by rhGM-CSF and rhIL-4 , CGL cells were differentiated into CGL-DCs. The phenotype of CGL-DC and the anti-tumor effects of cytotoxic T cells induced by CGL-DCs were investigated. The levels of IL-12 and IFN-γ in the supernatant were measured by ELISA. Results The DCs were successfully differenfiated from CGL cells. CGL cells were effectively and specifically destroyed by CTLs induced by CGL-DCs.Different levels of IL-12 were detected in the supernatant of cultured DCs and IFN-γ in the supernatant of cocultivated DCs and periphral blood mononuclear cells.Conclusion It has been confirmed that CGL-DC possesses immune function of antigen-presenting cell, in turn may have hopefully future in immunotherapy of CGL.
