应用ISSR标记研究新疆布顿大麦(Hordeum bogdanii Wilensky)的遗传多样性 被引量:1

摘要 利用35条ISSR引物对32份新疆布顿大麦(HordeumbogdaniiWilensky)的遗传多样性进行研究,发现仅9条ISSR引物能产生清晰扩增产物。在32份新疆布顿大麦中,9个ISSR标记共产生205条扩增片段,每条引物能产生12~45条,平均为22.8条。在这205条扩增片段中,有194条具有多态性,占94.6%,平均每个ISSR标记能产生21.6条多态性片段。ISSR标记揭示的32份新疆布顿大麦间的遗传相似系数变化范围为0.31~0.87,平均值为0.61。采用UPGMA法对32份新疆布顿大麦进行遗传聚类分析表明,ISSR标记能将32份新疆布顿大麦完全区分开,且ISSR标记揭示的新疆布顿大麦居群间的遗传关系与其地理来源间具有密切联系。 Using 35 ISSR primers,the genetic diversity among 32 accessions of hordeum bogdanii wilensky is investigated.Among 35 ISSR primers,9 primers can generate distinct amplified products.Among the 32 accessions of H.bogdanii,a total of 205 bands are detected by 9 primers,among which each primer amplifis 12 to 45 bands with the mean of 22.8 bands.One hundred and ninety-four out of 205 bands (94.6%) are polymorphic with 21.6 polymorphic bands per primer on average.The ISSR-based genetic similarity (GS) values among 32 H.bogdanii accessions ranges from 0.31 to 0.87,with the mean of 0.61.Based on the GS matrix,a dendrogram showing the genetic relationships between accessions is constructed by using the UPGMA method.The results show that all the 32 accessions can be distinguished by ISSR markers.It also indicates that the genetic relationships among H.bogdanii accessions based on ISSR markers is correlated with the geographical distribution.
