摘要
利用水煮醇沉法从桑叶中提取可溶性多糖,通过正交试验确定提取多糖的优化条件,用苯酚-硫酸法测定多糖含量,通过Seveg法除蛋白,粗多糖过葡聚糖凝胶(SephadexG-100)柱层析进行纯化分离,并用聚丙烯酰胺凝胶电泳进行纯度检查。研究结果表明,水浴温度100℃、桑叶∶水量=1∶100(w/v)、浸提60min、pH4为最佳提取条件,其多糖含量为98.52mg/g(DW)。柱层析纯化及聚丙烯酰胺凝胶电泳均表明桑叶多糖为单一多糖。
A water-soluble polysaccharide was extracted from Morus abla L. Leaves by method of boiling water and ethanol deposition. The optimal condition was determined by the orthogonal experiment. The content of polysaccharides was measured by phenol-sulfuric acid method. Through removing protein with Seveg and chromatography with Sephadex G-100 column, the crude polysaccharides were isolated and purified. And the purity was judged by polypropylene acylamide gelatin electrophoresis. The result shows that the optimal extraction condition is 100℃, 60 min, pH 4 as well as the ratio 1∶100(w/v). Under this condition 98.52mg/g(DW)polysaccharides was obtained. And the Morus abla L. Leaves polysaccharides were shown to be a homogenous component by the column chromatography and electrophoresis.
