摘要
应用脾内一次免疫法,按常规进行细胞融合,获得2株分泌抗鼠白细胞介素-4(IL-4)单克隆抗体(McAb))的杂交瘤细胞株。此杂交瘤分泌的McAb具有很强的特异性,对大肠杆菌(E.coli)表达的重组mIL-4呈阳性反应,与天然mIL-4呈阳性反应,而与E.coli蛋白、重组mIL-1、mIL-2、mIL-3及mIL-6无交叉反应。经Western印迹实验证实,此McAb可与IL-4表达带特异地结合,具有较强的封闭活性。
By means of primary intrasplenic immunization method, two rat monoclonal antibodies that are specific for mouse IL-4 have been produced. Anti-MrIL-4 McAb exhibits no cross reactivity to MrIL-1, MrIL-2, MrIL-3, MrIL-6 and E. coli protein. In addition, these McAb are capable of reacting with natural IL-4. Titre of media supernatant is 1:16. Antibodies to mouse IL-4 provide very efficient tools to research IL-4 in biological function and identify IL-4 in biological samples.
