摘要
探讨选择性环氧合酶 2 (COX 2 )抑制剂Celebrex对裸鼠胰腺癌PC 3细胞移植瘤生长和肿瘤组织新生血管生成的影响 ,为Celebrex的临床应用提供依据。本文采用体外观测Celebrex对移植瘤生长的影响 ,并选用Ⅳ型胶原作为肿瘤微血管标记物 ,测定移植瘤中微血管密度 (MVD)。结果显示 ,治疗组移植瘤生长曲线较对照组明显低平 ,对照组移植瘤近似体积为 0 4 38cm3 ,治疗组为 0 2 12cm3 (抑瘤率为 5 1 6 % ,P <0 0 5 )。对照组肿瘤组织MVD为 6 3 87± 13 6 7,治疗组 32 2 5± 12 99,两组间差异有非常显著性 (P <0 0 1)。表明COX 2可能参与了肿瘤新生血管的生成 ,选择性COX 2抑制剂Celebrex则具有抑制肿瘤生长和对抗肿瘤新生血管生成的作用。
To find the impact of Celebrex, a selective cyclooxygenase-2 inhibitor, on tumor growth and angiogenesis in pancreatic carcinoma of xenografted nude mice. The effects of Celebrex on tumor growth in vivo, and microvascular density (MVD) of tumors were measured by microvessel stained marker, collagen Ⅳ. The result showed MVD as 63.87±13.67 in control group while 32.25±12.99 in treated group. A significant correlation was observed between the two groups(P<0.01).It suggests that COX-2 may play an important role in the angiogenesis of pancreatic carcinoma and the selective COX-2 inhibitor, Celebrex, can inhibit angiogenesis and tumor growth.
