摘要 观察大鼠神经干细胞聚集体的内部结构并分析其细胞组成。培养神经干细胞 ,分别做HE染色 ;nestin、GFAP、NSE免疫组化 ;细胞超微结构应用扫描电镜和透射电镜观察。结果显示 ,细胞聚集体内有健康细胞和凋亡或坏死细胞 ,后者被前者包裹形成典型的鸟巢状结构。健康细胞分为两大类 ,一种是小细胞 ,电子密度低 ,细胞表面较光滑 ;nestin强阳性和 /或GFAP阳性。另一种细胞较大 ,有突起 ,电子密度高 ,nestin弱阳性。健康细胞间存在粘附连接 ,细胞内可见到吞噬体和板层状膜样结构。推测神经干细胞聚集体内的小细胞是较幼稚的神经干细胞 ,大细胞为处于分化前期的细胞。神经干细胞具有吞噬功能。 To investigate the structure of rat neural stem cell clusters in vitro and to analyze its cell types. The rat neural stem cells were cultured and microscopied with HE stainning and immunohistochemical stainning to detect nestin, GFAP and NSE antigen of the cells. The ultrastructure of the cells was observed under scanning and transmission electron microscope. It was noted that there were healthy cells and dead cells in the cell clusters. The healthy cells surrounded the dead cells to form a typical nest-like stucture. The healthy cells consisted of two cell types, small cell and large cell. The small cells were found to be with electron-lucent, smooth surface and nestin-positive. The large cells have electron-dense, many cell processes and weak nestin-positive. Healthy cells attached to each other by adherens junctions. Phagosomes and lamellar bodies were found in the healthy cells. It was suggested that the small cells in the clusters may belong to the immature neural stem cells, wherease the large cells could be the early differentiation cells. Neural stem cells showed phagocytic capability.
