摘要 目的:回顾神经生长因子的研究现状及其在周围神经损伤修复中的应用,明确其是否为治疗周围神经损伤的理想药物。资料来源:应用计算机检索“Medline1993-01/2004-05”,检索词“NerveGrowthFactor,并限定文章语言种类为English,并应用计算机检索CNKI中文期刊全文数据库,检索词“神经生长因子”,检索年份“2000/2004”限定文章语言为中文,检索词“神经生长因子”。资料选择:对PubMed检索到的资料进行初审,筛除明显不随机对照试验的研究,对剩余的文献手工检索查找全文。纳入标准为随机对照试验,并以相同标准筛选从CNKI中检索到的文章。共收集到符合标准的英文全文文献57篇,中文全文文献642篇。选择以动物为对象的文章,其中研究内容相似的,以近3年且发表在较权威杂志者优先的,排除综述类文献。数据提炼:将收集到的23篇文章,包括英文文献13篇,中文文献10篇,其中与神经生长因子的分子结构相关的2篇,与神经生长因子的分布、受体、生物学的作用相关的5篇,神经生长因子在周围神经损伤中的作用的16篇。资料综合:23篇文献包括725只动物,论述了神经生长因子的分子结构,神经生长因子的分布,生物学作用以及在周围神经损伤中的作用。结论:越来越多的证据表明给予外源性神经生长因子是帮助受损周围神经修复和再生的重要手? OBJECTIVE:To review the researches related to nerve growth factor (NGF) and its application in repairing peripheral nerve injury, and confirm whether it is the optimal agent for peripheral nerve injury renovation. DATA SOURCES: Using the key terms “nerve growth factor”, we searched the MEDLINE database in English language between January 1993 and May 2004.Meanwhile,using the same key terms in Chinese, we searched the CNKI database between 2002 and 2004. STUDY SELECTION:All articles searched from the PubMed database were selected firstly, and the investigations of obvious non randomized trial were excluded and the rest full text was looked up. The randomized controlled trials(RCT) were accepted as inclusive criteria. Meanwhile, all articles searched from the CNKI database were selected according to the same standards. A total of 45 English and 642 Chinese articles were collected. The articles with animals as subjects and published in more authoritative journal were collected firstly. The articles of summarization were excluded. DATA EXTRACTION: A total of 23 articles were collected,13 articles were English, and 10 articles were Chinese. Among them 2 articles were related to the molecular constitution of NGF, 5 articles related to the distribution, receptor and biological function of NGF and 16 articles related to the function of NGF on peripheral nerve injury. DATA SYNTHESIS: The collected 23 articles involved 725 animals. The molecular constitution, distribution, and biological function of NGF and the function of NGF on peripheral nerve injury were discussed. CONCLUSION:More and more evidences indicate that exogenous NGF is the important method for the repair and regeneration of impaired peripheral nerves. Large scale clinical trials and the administration pathway of exogenous NGF should be the major investing direction.
